摘要
目的:观察高糖培养条件下大鼠红细胞多元醇代谢及脂质氧化的变化及二甲双胍对此的干预作用。方法:用高糖培养大鼠红细胞,测定大鼠红细胞中山梨醇的含量及培养上清液中一氧化氮(NO)含量的变化,并观察加入不同浓度二甲双胍(2×10-5、4×10-6、8×10-7 mol.L-1)处理后对山梨醇和NO含量的影响,同时设立低糖培养的正常对照组进行比较。结果:与正常对照组比较,高糖对照组大鼠山梨醇含量升高、NO含量降低(P<0.01);与高糖对照组比较,加入二甲双胍后能显著降低山梨醇含量并可升高NO的含量(P<0.01或P<0.05)。结论:二甲双胍可通过抑制山梨醇含量的升高及增加NO的水平减轻高糖所致的红细胞损伤。
OBJECTIVE:To observe the changes of lipid oxidation and polyol pathway metabolism of rat erythrocytes cultured in high glucose medium and intervention effect of metformin.METHODS:Rat erythrocytes were cultured in high glucose medium and the content of sorbitol in erythrocytes and the content of NO in culture medium were determined.The effect of metformin at different concentrations(2×10^-5 mol·L^-1,4×10^-6 mol·L^-1,8×10^-7 mol·L^-1)on the level of sorbitol and NO were studied with rat erythrocytes cultured in low glucose medium as normal control.RESULTS:Compared with normal control group,the level of sorbitol in high glucose group was increased while the content of NO was decreased(P〈0.01).Compared with high glucose group,metformin groups had lower level of sorbitol and higher level of NO(P〈0.01 or P〈0.05).CONCLUSION:Metformin can relieve injury of erythrocytes induced by high glucose by inhibiting the increase of sorbitol and decrease of NO.
