摘要
目的探讨山楂叶总黄酮(flavone mixture of crataegus leaves,FMCL)对H2O2诱导的PC12细胞凋亡的保护作用及其分子机制。方法用H2O2刺激PC12细胞使其发生凋亡,用流式细胞仪检测PC12细胞凋亡率,用Westernblot法检测Bcl-2、Bax、Caspase-3P20蛋白及用RT-PCR法检测Bcl-2、Bax mRNA表达量的变化。结果与模型组比较FMCL 100μg/ml剂量组PC12细胞凋亡率下降,Bcl-2蛋白及mRNA表达量明显升高,而Bax、Caspase-3P20蛋白及BaxmRNA表达量明显降低(P<0.05或P<0.01),且在一定范围内存在剂量依赖关系。结论山楂叶总黄酮可抑制H2O2诱导的PC12细胞凋亡,其机制可能与调节凋亡相关基因及蛋白的表达从而抑制Caspase-3活化有关。
Objective To discuss the mechanism of the protective effect of FMCL on apoptosis of PC12 cells induced by H2O2 .Methods Based on the model of apoptosis of PC12 cells induced by H2O2,apoptosis rate was analyzed by flow cytometer. The expression of mRNA and protein were tested by RT-PCR and Western blot. Results The apoptosis rate was decreased in the FMCL treatment group. The mRNA and protein expression of Bcl-2 were increased and that of Bax was decreased( P 〈 0.05 or P 〈 0.01 ). Thus the ratio of Bcl-2/ Bax increased. The activity fragment of caspase-3P20 was decreased( P 〈 0.05). Conclusion FMCL can prevent PC12 cells against H2O2 oxidation damage. The mechanism of protection is likely related to increasing the ratio of Bcl-2/Bax so that can decrease the activation of easpase-3.
