摘要
目的采用毛细管区带电泳法(CZE)测定血浆中头孢羟氨苄、甲氧苄啶的含量。方法以20mmol/L硼砂(pH=7.5)为背景电解质,35cm×75μm(i.d)未涂层毛细管为色谱柱,于267nm处检测,操作温度为25℃,分离电压为20kV,采用峰面积内标法定量。结果头孢羟氨苄和甲氧苄啶的线性范围分别为0.4—4.0μg/mL,0.25—4.0μg/mL,该方法测得头孢羟氨苄和甲氧苄啶在低、中、高浓度下的回收率均在97%以上,迁移时间的RSD分别为1.23%,2.41%;峰面积的RSD分别为2.23%,1.60%,日内、日间精密度均符合方法学要求。结论该方法用量少,简便,快速,准确,可作为该药品生物等效性研究中的有效分析方法。
OBJECTIVE A new capillary zone electrophoretic method has been established for the rapid determination of cefadroxil and trimethoprim in plasma. METHODS The analysis was performed in an uncoated fsedsilica capillary with 75μm( i. d). And total length socm with a buffer solution of 20mmol/L disodium tetraborate ( pH7.5 ). The applied voltage was 20kV and UV detection wavelength was 267nm,intemal calibration curve of peak area was selected. RESULTS The linear range of cefadroxil and trimethoprim were0.4 -4.0μg/mL,0.25 - 4.0μg/mL,respeetively. The recoveries obtained from plasma for the two medicine were more than 97%. The intra-day and inter-day presision assessed at three mass concentration levels were satisfactory. The relative standard deviation(RSD) value for migration time of cefadroxil and trimethoprim were 1.23% and 2.41% ,respectively;the RSD value for peak area was 2.23% and 1.60%. CONCLUSION The method is less solvent consumption,simple,rapid and accurate,and it can be applied to the research biological equivalent.
