摘要
目的:研究胆囊癌组织中IL-8mRNA和TSP-1mRNA表达及其对肿瘤血管生成的作用。方法:52例胆囊癌手术切除标本经4%甲醛固定后常规制作石蜡包埋切片,采用CD34单克隆抗体和免疫组化ABC法显示肿瘤组织中微血管,采用原位杂交技术观察肿瘤细胞中IL-8mR-NA和TSP-1mRNA的表达。结果:52例胆囊癌组织中IL-8mRNA和TSP-1mR-NA阳性率分别为67·3%(35/52)和59·6%(31/52),微血管计数(microvesselcount,MVC)为62·4±12·9个/HP。高分化腺癌和未转移病例MVC(52·4±17·5;53·6±19·3)和IL-8mRNA阳性率(44·0%,11/25;53·6%,15/28)明显低于中低分化腺癌(71·5±18·3,P<0·01;87·5%,14/18,P<0·05)或未分化癌(86·2±23·4,P<0·01;100·0%,5/5,P=0·03)及转移病例(73·4±16·8,P<0·01;83·3%,20/24,P<0·05),但TSP-1mRNA表达则相反。IL-8mRNA阳性病例和TSP-1mRNA阴性病例MVC(68·8±15·4;69·5±19·2)明显高于IL-8mRNA阴性病例(49·2±18·7,P<0·01)和TSP-1mRNA阳性病例(57·7±16·3,P<0·01)。IL-8mRNA与TSP-1mRNA表达呈密切相关,P<0·01。结论:IL-8mRNA和TSP-1mRNA表达与胆囊癌血管生成密切相关,IL-8具有促血管生成作用,而TSP-1具有抑血管生成作用。
OBJECTIVE:To study the expressions of IL-8 mRNA and TSP-1 mRNA and their effects on the promoting or inhibiting angiogenesis of gallbladder carcinoma tissues. METHODS: The assays of IL-8 mRNA and TSP-1 mRNA were performed by in situ hybridization techniques and the microvessel counts (MVC) were completed by the immunohistochmical method of avidin-biotin complex on the 10 % neutral formalin-fixed and routinely paraffin-embedded sections of 52 cases of gallbladder carcinoma. RESULTS: The positive ratse of IL-8 mRNA and TSP-1 mRNA were 67. 3 % (35/52) and 59.6% (31/52), respectively. The MVC was 62.4±12.9/HP. The positive rates of IL-8 mRNA were significantly lower in the well-differentiated (44.0%, 11/25) and without metastasitic cases (53.6%, 15/28) than those in the middle-t poorly-differentiated (87.5%, 14/18, P〉0. 05), un-differentiated (100%, 5/5, P= 0. 03) and with metastastic ones (83.3%, 20/24, P〈 0. 05), but that of TSP-1 mRNA was opposite. The MVCs were significantly lower in well differentiated (55.2 ±17.5) and without-metastastic cases (53.6 ± 19.3) than those in the middle-t poorly-differentiated (71.5±18.3,P〈0.01) or un-differentiated (86.2±23. 4,P〈0.01) and with metastastic ones (73.4 ± 16.8,P〈0. 01). The MVCs were significantly higher in the positive cases of IL-8 mRNA (68.8 ± 15.4) or negative cases of TSP-1 mRNA (69.5±19.2) than those of the negative ones of IL-8 mRNA (49.2±18.7, P〈0.01) or positive ones of TSP-1 mRNA (57.7±16.3,P〈0.01). The closely negative correlation was found between the expressions of IL-8 mRNA and TSP-1 mRNA (P〈0. 01). CONCLUSION: The expressions of IL-8 mRNA and TSP-1 mRNA might he closely related to the angiogenesis of gallbladder carcinoma tissues, IL-8 might have the promoting effect and TSP-1 might have inhibiting effect on the angiogenesis of malignant lesions.
