摘要
目的研究ATP受体P2X2、P2X3在大鼠脊神经节感觉神经元的表达、形态特点及与神经肽降钙素基因相关肽(CGRP)、钙结合蛋白calretinin的共存。方法应用激光共聚焦显微镜结合免疫组织化学双标技术。结果免疫组织化学显示脊神经节内可见大量P2X2、P2X3受体阳性的细胞和神经纤维;P2X2受体亚型阳性细胞多为大、中、小型神经元,P2X3受体亚型阳性细胞为中、小型神经元;免疫组织化学双标显示有41.2±1.9%P2X2受体阳性细胞含有CGRP,而P2X3受体亚型与CGRP共存的细胞很难见到;有8.9±1.1%的P2X2免疫阳性神经元和7.2±1.3%P2X3受体阳性神经元分别含有calretinin。结论这一结果表明大鼠脊神经节内表达的ATP受体P2X2、P2X3是不同的受体亚型。
Objective: To study the expression of purinergic receptor, P2X2 and P2X3 subunites in sensory neurons of the rat dorsal root ganglion, their morphology and co - localization with neuropeptide, calcitonin gene - related peptide (CGRP) and calcium binding- protein, calretinin. Methods: -Laser scanning confocal microscope combined with immunohistochemical double labeling methods were used. Results: Most of P2X2 and P2X3 imrnunoreactive sensory neurons and fibers were observed in the dorsal root ganglion. The expression of P2X2 receptor was found in large-, middle- and small -sized cell bodies and P2X3 receptor in middle - and small - sized neurons. Immunohistoehemieal double - labeling showed that about 41.2 ± 1.9% P2X2 immunoreactive neurons co- stained with CGRP immunoreaetivity, but co- local- ization of P2X3 receptor with CGRP in sensory cell bodies was very few. There were 8. 9 ± 1.1% P2X2 immunopositive neurons and 7.2 ± 1. 3% P2X3 iremunoreaetive neurons contained ealretinin respeetivelyo Conclusion: These results demonstrated that P2X2 and P2X3 were different receptor subunites in sensory neurons of rat dorsal root ganglion.
