回转器模拟失重对人胃癌SGC-7901细胞形态和生物学特征的影响 被引量:5

摘要 目的探讨模拟失重对人胃癌SGC-7901细胞形态、增殖、周期和凋亡的影响。方法采用回转器模拟失重,用图像分析系统软件分析HE染色后SGC-7901细胞形态的改变,用免疫组化法观察细胞增殖细胞核抗原(PCNA)表达的变化,用流式细胞仪测定细胞周期,并用TIJNEL法检测细胞凋亡。结果回转器模拟失重后,回转组SGC-7901细胞12 h、24 h、36 h、48 h及72 h各时间段的细胞面积均较对照组缩小(P<0.01);在36 h时,其长短径之比较对照组增大(P<0.01),其余时间段无明显差异。12 h、24 h和36 h回转组SGC-7901细胞PCNA抗体阳性细胞比率与对照组相比无明显差异,48 h、72 h回转组的PCNA抗体阳性细胞明显减少(P<0.05或P<0.01)。12 h、 24 h和36 h回转组SGC-7901细胞的细胞周期与对照组相比无明显差异,48 h、72 h回转后G0+G1 期细胞显著增多(P<0.01或P<0.05),S+G2+M期细胞则显著减少(P<0.01或P<0.05)。 12 h、24 h、36 h和48 h回转组SGC-7901细胞凋亡比例与对照组相比无明显差异,72 h回转组细胞凋亡比例较对照组明显增多(P<0.01)。结论回转器模拟失重使胃癌SGC-7901细胞的形态发生了变化,48 h、72 h时出现细胞周期阻滞、增殖抑制,72 h时凋亡增加。 Objective To investigate the effects of simulated weightlessness on cellular morphology, proliferation, cell cycle and apoptosis of human gastric carcinoma cell line SGC-7901. Methods Five experimental groups of the cell line were cultured in the rotating clinostat for 12 h, 24 h, 36 h, 48 h and 72 h respectively. Image-pro4.5 image analysis system was employed for morphometric study. The expressions of proliferating cell nuclear antigen (PCNA) were examined by immunohistochemical stain. The changes of cell cycle were examined by a cytometer. Apoptosis was measured by TUNEL method. Results Compared with control group, the area of cell line SGC-7901 of all 5 experimental groups decreased significantly (P〈0. 01), the ratio of long diameter and short diameter increased significantly in 36 h simulated weightlessness group(P〈0.01),but did not change significantly in other groups. Compared with control, the expression of PCNA of cell line SGC-7901 did not change significantly in 12 h, 24 h, and 36 h groups, but decreased significantly in 48 h and 72 h groups (P〈0.05 or P〈0.01). Compared with control, the cell cycles of cell line SGC-7901 in 12 h, 24 h, and 36 h groups did not change significantly, but the cells in Go+G1 phase in 48 h and 72 h groups increased significantly (P〈0.01 or P〈0. 05) ; accordingly, the cells in S+G2 +M phase decreased significantly (P〈0.0l or P〈0. 05). Compared with control, the apoptosis index of cell line SGC-7901 in 12 h, 24 h, 36 h and 48 h groups did not change significantly, but increased significantly in 72 h group. Conclusions Simulated weightlessness in a rotating clinostat may lead morphological change of human carcinoma cell line. The conversion of cell from G1 phase to S phase was blocked, the proliferation was inhibited after rotating for 48 h and 72 h, the increased apoptosis index was found after 72 h rotating.
