摘要
目的比较1型和4型HEVORF2抗原性的差异。方法对1型和4型ORF2同样位置各3个抗原片段分别进行表达,1型ORF2的3个重叠抗原以从N端到C端的顺序编号为1pET1、1pET2和1pET3;同样4型3个片段的编号为4pET1、4pET2和4pET3。用纯化后的各抗原片段分别建立酶联免疫检测方法检测临床肝炎病人血清136份,对检测结果进行统计分析。结果对6个抗原片段成功地进行了表达,纯化后6个抗原片段的纯度在95%以上。通过检测136份临床肝炎病人血清,显示1型抗原中3个抗原片段均存在抗原性差异,4型病毒ORF2中间的抗原片段与C端和N端抗原片段存在抗原性差异,1型和4型病毒之间N端抗原存在抗原性差异,具有统计学意义。结论1型和4型HEVORF2N端重组蛋白之间存在抗原性差异。
Objective To compare ORF 2 antigenicity between HEV genotypes 1 and 4. Methods Three overlapping fragments at corresponding position on ORF2 of genotypes 1 and 4 which cover the full-length of ORF2 were cloned and expressed in E. coli system. The expressed polypeptides were purified. The ELISA for each fragment was developed with the corresponding purified polypeptide. One hundred and thirty six serum samples from patients, was tested. The detecting results with different ELISA method were compared and analyzed. The six fragments were successfully expressed in E. coli system. The purity of the six purified polypeptides was more than 95%. The results showed that three polypeptides from genotype 1 ORF2 were significantly diffenmt, the middle polypeptide from genotype 4 ORF2 was significantly different from fragments of N and C temurms, the fragments at N tennirms from genotypes 1 and 4 were significantly different. Conclusion Antigenicity difference exists at N tennirms of genotypes 1 and 4 ORF2.
