母体免疫球蛋白G对N-甲基-D-天冬氨酸诱导婴儿痉挛大鼠模型的作用及其对脑内促肾上腺皮质激素表达的影响 被引量:3

摘要 目的探讨母体免疫球蛋白G(IgG)对N-甲基-D-天冬氨酸(NMDA)诱导婴儿痉挛大鼠模型的临床作用及其对脑内促肾上腺皮质激素(ACTH)免疫反应阳性细胞表达影响。方法幼鼠36只,出生后即行母子分离,随机分为对照组、NMDA组、母体IgG干预组。根据发作程度,对NMDA致症状进行评分,并对NMDA诱导抱团发作潜伏期分布情况进行比较,采用免疫组织化学法,观察各组实验动物脑神经细胞ACTH表达变化。结果1.母体IgG干预组症状评分(3.53)低于NMDA组(5.67),差异具有显著性(t=2.695 P=0.012)。2.母体IgG干预组抱团发作潜伏期≥40 min的比率(80%)明显高于NMDA组(33%)(P=0.022)。3.在顶区和颞区皮质3组ACTH免疫阳性细胞数目间分别存在着显著性差异,阳性细胞计数由多到少依次为母体IgG干预组>NMDA组>对照组。在丘脑,只有对照组和母体IgG干预组之间存在显著性差异。结论1.母体IgG对于婴儿痉挛大鼠模型具有控制痉挛作用。2.ACTH对中枢神经系统存在直接作用,母体IgG预处理后,脑内ACTH免疫反应阳性细胞增加,推测母体IgG可能部分通过ACTH发挥神经系统保护作用。 Objective To explore the effect of maternal immunoglobulin G(IgG) on N-methyl-D-aspartate(NMDA) inducecl infantile spasms rat model and on the adrenocortieotrophic hormone(ACTH) expression in brain. Methods Thirty-six neonatal rats separated from their mothers were randomly divided into 3 groups: control group ( n = 9) ;NMDA group( n = 12) ;maternal IgG group( n=15). Clinical seizure grading and emprosthotonus latent period evaluation was carried out. Expressions of ACTH immuno-reactive cells were also observed in brain in different groups. Results 1. The average seizure grade of maternal IgG group (3.53) was significantly lower than that of NMDA group (5.67), t = 2. 695 P = 0. 012.2. The ratio of emprosthotonus latent period which was longer than 40 minutes in maternal IgG group(80%) was significantly higher than that in NMDA group(33%)( P = 0. 022). 3. The counts of ACTH immuno-reactive cells were significantly different from other 2 different groups both in vertex and temporal cortex(maternal IgG group〉 MDA group〉control group). In thalamencephalon,there existed significance only between control group and maternal IgG group. Conclusions 1. The maternal IgG pretreatrnent controls NMDA- induced spasm and prolonged the ernprosthotonus latent period. 2, The maternal IgG pretreatment can increase the counts of ACTH immuno-reactive cells in brain in infantile spasm rat model,which suggests that maternal IgG may protect central nerve system partly by elevating ACTH concentration in brain.
