压力超负荷性心肌肥厚大鼠血浆ADM和ET变化的实验研究

Experimental Study of Changes of Plasma Adrenomedulin and Endothelin in Pressure Overload Rat with Hypertrophy

摘要 目的:探讨血浆肾上腺髓质素(ADM)在大鼠压力超负荷性心肌肥厚中的病理生理作用及与内皮素(ET)的关系。方法:20只Wistar大鼠随机分为假手术组和腹主动脉部分缩窄致心肌肥厚组。均在术后6周观察尾动脉收缩压(SBP),左心系数(左心室重/体重)的变化,采用放免分析法检测血浆ADM和ET水平。结果:心肌肥厚组与假手术组比较,SBP、左心系数分别增加161.7%和119.3%(P<0.01),血浆ADM、ET及ET/ADM值分别增加158.5%、210.9%、131.5%(P<0.01),ADM、ET与左心系数呈正相关,ADM与ET呈正相关。结论:在此心肌肥厚模型中肾上腺髓质素作为循环激素被活化,可能是一种代偿性升高。心肌肥厚时ET、ADM间的动态平衡被打破,其调节紊乱可能是发病的重要原因之一。 Objective: To investigate the pathophysiologic effect of plasma adrenomedullin (ADM) and its relation to endothelin (ET) in pressure overload rats with hypertrophy. Methods:20 wistar rats were randomly divided into two groups:sham- operation group and partial abdominal arota constriction group. The changes of systolic blood pressure (SBP) and left ventricular mess index (left ventricular weight/body weight) were observed at six weeks after operation. The levels of ADM and ET were determined by radioimmunoassay method. Results:Compared with sham- operation group,SBP and left ventricular mess index in partial abdominal arota constriction group was increased by 161.7 % and 119.3 %, respectively (P 〈 0.01), plasma ADM, ET and ratio of ET/ADM was increased by 158.5 % ,210.9 % and 131.5 % , respectively (P 〈 0.01). There was positive correlation between ADM, ET and left ventricular mess index,or ADM and ET. Conclusion:ADM is activated as a circulation hormone in thy hypertrophied model. The increasing ADM may be a compensating reaction. The disbalance between ADM and ET and disordered regulation may be important etiological factors.
