摘要
目的:对顺铂前体脂质体制备工艺及包封率测定方法进行初步研究。方法:选用山梨醇为水溶性载体,采用前体脂质体法与超声分散混合法制备顺铂前体脂质体;高效液相色谱法测得包封率。结果:制得的顺铂前体脂质体为球形双层脂质体,包封率为71.8%。结论:本法制备的顺铂前体脂质体,在水化前便于贮存,重新混悬时易于分散、工艺简单、可规模化生产、包封率高;高效液相色谱法测定顺铂的含量,方法简便、准确。
OBJECTIVE To Study the preparation of pro-liposomes of cisplatin and determination method of encapsulation percentage. METHODS Cisplatin liposomes were prepared by the technology of pro-liposome and sonication, using sorbitol as water-solubility carrier, and encapsulation percentage was determined by HPLC. RESULTS The cisplatin liposomes were double-layer sphericity, the average encapsulation percentage was 71.8%. CONCLUSION The pro-liposomes can be easily reserved before hydrated, and could be well dispersed when suspended in water. The technology is simple and convenient for industrial production, and a relatively high encapsulation percentage can be obtained;The determination method of HPLC is simple and convenient for precise.
