摘要
目的:建立高效液相色谱(HPLC)法检测人血浆中9-硝基喜树碱的浓度。方法:采用Kromasil C18色谱分析柱,以甲醇-磷酸盐缓冲液(48:52,pH 2.5)作为流动相,流速为1.0mL·min^-1,柱温30℃,检测波长370nm,喜树碱为内标,血浆样品用C18固相萃取小柱处理,采用加权最小二乘法计算回归方程。结果:9-硝基喜树碱的线性范围为5~100μg·L^-1,r=0.9999,最低检测限为5μg·L^-1。该方法的提取回收率为83.5%~114.4%,批间和批内RSD〈15%。结论:该方法简便、准确、重现性好且灵敏度高,适用于9-硝基喜树碱临床人体药动学研究。
OBJECTIVE To establish an HPLC method for the determination of 9-nitro-camptothecin in human plasma. METHODS A kromasil C18 column was used as analytical column. The mobile phase consisted of methanol and phosphate buffer solution(48:52,pH 2. 5). The flow rate was 1.0 mL·min^-1 and the column temprature was at 30℃. Detection wavelength was at 3711 nm. Camptothecin was used as internal standard. Plasma samples were prepared with solid phase extraction assay. RESULTS A linear calibration curve of 9-nitro-camptothecin in plasma was obtained in the concentration range of 5 to 1000μg·L^-1(r= 0. 999 9). The absolute recovery was between 83.5%-114. 4% and the relative recovery is between 84. 0% - 104. 9%. The intra- and inter-run precision was measured to be below 15 %. CONCLUSION The method is shown to be convenient and accurate,and suitable for clinical pharmacokinetic study of 9-nitro-camptothecin.
