运动心脏重塑过程中降钙素基因相关肽基因的表达 被引量:7

Gene Expression of Calcitonin Gene Related Peptide in Rat's Heart during Exercise-Induced Cardiac Remodeling

摘要 目的:从基因水平上探讨运动性心脏重塑的发生机制。方法:70只SD大鼠随机分为对照组和实验组,实验组进行75天的跑台耐力训练。分别于训练第3、10、23、37和75天次日晨宰杀实验组大鼠,每次7只。相应对照组亦于当天上午取材。以异硫氰酸胍法提取大鼠心室肌组织总RNA,聚合酶链式反应技术(PCR)测定心室肌降钙素基因相关肽(Calcitonin Gene Related Peptide,CGRP)mRNA的表达量,β-actin为内参照,CGRP mRNA/β-actin的比值为每个测试样本中CGRPmRNA的表达值。结果:在5个时相点中,实验组大鼠心室肌组织CGRP mRNA表达量与对照组相比无显著差异。结论:运动性心脏重塑过程中,大鼠心室肌组织CGRP mRNA表达量无明显变化。 In order to further investigate the molecular mechanism of exercise - induced cardiac remodeling, the dynamic changes in calcitonin gene related peptide(CGRP) mRNA expression in ventricle were studied. Seventy rats, receiving food and water at/labium, were randomly divided into two groups: control group and trained group (used as the model of physiological cardial hypertrophy). Rats in training group exercised on treadmill 6 days per week. For the first day, the speed of treadmill was set at 15m/min, then the duration was gradually increased from 15min/day, to 60min/day, and intensity increased from 15m/min to 30m/min in a period of a week. Thereafter, the rats in training group continued training for 75 days. CGRP mRNA expression in ventricle was detected at the 3rd day, 10th day, 23rd day, 37th day and 75th day, respectively by RT - PCR. The results showed that the expression of ventriular CGRP mRNA revealed no significant difference from the 3rd day to the 75th day, suggesting that CGRP mRNA expression in ventricle remained unchanged during exercise - induced cardiac remodeling and that CGRP did not participate in this remodeling process.
