摘要 以毒三素链霉菌(stretomyces toxytricini)突变株XC-lp-69为试验菌株,研究了利普司他汀(lipstatin)的发酵工艺.通过单因素条件试验确定了其生物合成的最适温度,种龄,接种量及摇瓶装量.以正交试验优化了其发酵培养基,并进行了发酵中间补料试验,确定了最佳补料工艺.试验结果表明:以5.5%豆油,2.0%甘油,3.0%黄豆粉,1.0%酵母粉,1.2%卵磷脂的发酵培养基配方,以20h种龄,接种量2.0%,在250 mL三角瓶中装量25 mL,27℃下发酵,在发酵46 h时补加豆油/甘油混合物,利普司他汀的发酵效价达1 400μg.mL-1. The fermentation process by Streptomyces toxytricini XC-lp-69 for increasing the productivity of Lipstatin was studied. Through single-factor experiments, the optimum fermentation temperature, seed age, inoculation volume and loading amount were obtained. The fermentation medium composition was investigated by orthogonal experiments. The titer of Lipstatin increased greatly by using feeding fermentation. The result showed that the titer of Lipstatin reached 1400μg·mL^-1 with 20 h seed age and 2.0% inoculation volume at 27℃ in 250 mL flake in which 25 mL medium of 5.5% bean oil, 2.0% glycerin, 3. 0% soybean powder, 1.0% yeast powder and 1.2% lecithin were loaded, and the bean oil/glycerin mixture was fed after it had been fermentated for 46 h.
