摘要
目的观察清脂益肝汤和二甲双胍治疗脂肪肝大鼠在疗效、生化、病理、脂肪代谢、胰岛素及瘦素基因表达各方面指标的变化。方法雄性Wistar大鼠43只,按体重层次随机分为4组,对照组(Ⅰ组)10只、模型组(Ⅱ组)11只、清脂益肝汤组(Ⅲ组)11只、二甲双胍组(Ⅳ组)11只,对照组饲以普通鼠饲料,其余三组均饲以高脂饲料。饲养4周后病理证实脂肪肝己形成,随后进行灌胃治疗,Ⅲ组16g生药·kg-1·d-1,Ⅳ组二甲双胍170mg·kg-1·d-1,每天一次5m1灌胃,Ⅰ组及Ⅱ组大鼠灌服等量蒸馏水,各组按原饲料进行饲养。8周后,全部处死,取血及肝组织,测定血中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、甘油三酯(TG)、胆固醇(CHO)、空腹胰岛素(FINS)、空腹血糖(FBS)、游离脂肪酸(FFA),瘦素m-RNA(Leptinm-RNA)在肝组织内表达,观察肝脏组织学变化。结果脂肪肝大鼠存在明显脂肪代谢紊乱,肝功能异常,胰岛素抵抗及瘦素m-RNA表达增高;清脂益肝汤和二甲双胍能显著改善肝功能、降低血脂、降低瘦素高表达、改善胰岛素抵抗及肝组织学病理。结论清脂益肝汤和二甲双胍对实验性大鼠脂肪肝有一定治疗效果。
Objective To observe the change of the liver function, histology, fatty metabolism, insulin, leptin m-RNA and so on,after using mefformin and Qingzhiyigan decoction in rats with non-alcohol fatty liver.Methods 43 male Wistar rats were randomly divided into four groups, control group ( n = 10),model group ( n = 11),metformin group ( n = 11),Qingzhiyigan decoction group ( n = 11). The rats in control group were fed with normal diet,and the rats in other three groups were fed with a high fat diet. After four weeks,fatty liver model was established. The rats in model group,metformin group and Qingzhiyigan decoction group were fed with high fat diet while the rats in control group were fed with normal diet. After eight weeks all the animals were killed. Serum activities of liver-associated enzymes, blood lipids,FFA,leptin m-RNA,fasting blood glucose,fasting plasma insulin, HOMA insulin resistance index (HOMAIR) and the liver histology of rats in all groups were detected.Results The serum ALT,ASF,TC,TG,HOMA-IR,FFA and leptin m-RNA in metformin group and Qingzhiyigan decoction group were significantly lower than those in model group . The liver histology in the metformin group and Qingzhiyigan decoction group was improved significantly. Qingzhiyigan decoction and metformin may be effective in treating rat fatty liver induced by high fat diet.
