水溶性苦瓜多糖的分离纯化及生物学活性的研究 被引量:24

摘要 本文建立了苦瓜水溶性多糖的提取纯化方法,并对其理化性质及生物学活性进行检测。采用双酶法得到的苦瓜水溶性多糖,通过正交试验确定其最佳水解条件为:纤维素酶-提取温度50℃,pH值6.0,酶用量5%;中性蛋白酶-水解时间48h,pH值7.0,酶用量10%。多糖粗品经DEAE-32、SephadexG-75柱层析得到单一组分,命名为PMCⅠ。PMCⅠ的酸性水解液经薄层层析分析,其单糖组分别为葡萄糖、半乳糖、甘露糖和鼠李糖。对PMCⅠ生物功能的研究表明,其对羟基自由基、超氧阴离子自由基均有清除作用,IC50分别为0.0017mg/ml和0.005mg/ml,最大抑制率可达100%,证实其具有抗氧化的活性。抑菌实验表明其对金黄色葡萄球菌、大肠杆菌、鼠伤寒沙门氏菌、枯草杆菌等病原菌均有抑制作用,IC50分别为0.10、0.15、0.13、0.13mg/ml,其中以金黄色葡萄球菌最为明显。 The method of extracting and purifying polysaccharide from Momordica charantia was suggested in this paper. Its physical and chemical characteristics and biological activity were determined. The polysaccharide was extracted from Momordica charantia by adding two species of enzymes. The optimum hydrolysis conditions were got. The polysaccharide was further purified with DEAE-32 column chromatography and Sephadex G-75 column chromatography respectively until the single peak was got, and named as PMC Ⅰ. The acidic hydrolysis solution of PMC Ⅰ was analysed by means of TLC: its constituents respectively glucose, galactose, mannose, rhanmose. The study for its biological activity showed that it could eliminate free radicals such as · OH and O2^-·, and its IC50 for · OH and O2^- · were respectively 0.0017mg/ml and 0.005mg/ml. So it showed the activity of antioxidant. The experiments of inhibiting germs show that PMC Ⅰ can inhibit germs such as S.Aureus, E.coli, B.subtilus and S.typhimurium. The IC50 for those were respectively 0.10, 0.15, 0.13, and 0.13mg/ml, the most remarkable of which was S.Aureus.
