摘要
目的研究比较人参不同部位(全草、根及茎叶)提取的人参皂苷对犬急性心肌缺血的保护作用。方法采用结扎冠状动脉前降支的方法,造成犬急性实验性心肌缺血模型;描记心外膜电图,计算心肌缺血程度;用硝基蓝四唑(N-BT)进行心肌组织化学染色测定心肌梗死面积;用放射免疫法测定6-酮前列腺素F1α(6-Keto-PGF1α)与血栓素B2(TXB2)的比值;同时测定冠脉血流量。结果3种部位人参皂苷均具有显著改善犬急性心肌缺血的作用,在减轻心肌缺血程度,增加冠脉血流量等方面,作用强度基本相当。但人参全草提取的皂苷在减小心肌梗死面积,增加6-Keto-PGF1α/TXB2的比值方面较人参根提取皂苷、人参茎叶提取皂苷作用强。根和茎叶提取的皂苷作用无显著性差异。结论3种不同部位的人参皂苷均有明显对抗犬急性心肌缺血的作用,但药物作用强度有所不同。
Objective To compare the protective effects of ginsenosides extracted from different parts (including the whole plant, root, stem and leaves) of ginseng on acute myocardial ischemia in dogs. Methods The acute ischemic models of dog were set up by ligation of left ventricular anterior artery (LAD). The epicardiogram mapping was used to measure the degree of myocardial ischemia, quantitative histological assay (nitroblue tetrazolium, N -BT stain) was used to determine the area of myocardial infarction, and radioimmunological assay Was used to measure the changes of the ratio of 6 - keto - prostaglandin F1α/and thromboxane B2 (6 - Keto - PGF1α/TXB2) in serum. Meanwhile the coronary blood flow was determined. Results Ginsenosides from three parts of ginseng could obviously alleviate damages caused by myocardial ischemia and had similar effect in relieving myocardial ischemia and increasing the coronary blood flow. However, ginsenoside from the whole plant has a stronger effect in reducing the ischemic area and increasing the ratio of 6 - Keto - PGF1α/ TXB2 than that from root or leaves. And there was no significant difference between ginsenoside from root and ginsenoside from leaves. Conclusion Ginsenosides from the whole part, root and leaves of ginseng have obvious inhibition on the damages caused by myocardial ischemia, the effect being different potency.
