摘要
目的探讨不同浓度金钠多对由缺氧所致培养的血管内皮细胞分泌内皮素功能的影响.方法将培养的血管内皮细胞分为空白对照组、单纯缺氧组、缺氧加高浓度金钠多组、缺氧加中浓度金钠多组和缺氧加低浓度金钠多组.采用低压舱上升至5000m高度、停留30min,对培养的血管内皮细胞进行缺氧.用放射免疫法测定缺氧前和缺氧后0.5,6,24h各组细胞培养液中内皮素含量.结果①急性缺氧可显著促进血管内皮细胞分泌内皮素,缺氧后0.5h内皮素含量即显著升高,于缺氧后6h分泌内皮素含量最高.②金钠多能明显抑制缺氧促血管内皮细胞分泌内皮素的作用,在缺氧后0.5h表现出高浓度金钠多强于中浓度和低浓度金钠多组,在缺氧后6和24h则中浓度金钠多的作用要强于高浓度和低浓度组.结论金钠多能显著抑制缺氧促血管内皮细胞分泌内皮素的作用,且中浓度金钠多较高浓度和低浓度金钠多作用更明显.
AIM: To explore the effects of Ginaton at different concentrations on the hypoxia-induced endothelin (ET) secretion from cultured vascular endothelial cells (VECs). METHODS : Cultured VECs were divided into 5 groups: control group, simple hypoxia group, high-concentration Ginaton group, moderate-concentration Ginaton group, and low-concentration Ginaton group. For hypoxia, all groups except control group were put in a hypobaric chamber at the height of 5000 m for 30 min. The ET contents in supernatant of VECs before hypoxia and 0. 5 h after hypoxia, 6 h after hypoxia as well as 24 h after hypoxia were measured with radioimmunoassay. RESULTS: ① Simple hypoxia promoted VECs' secretion of ET and ET content increased significantly at 0.5 h after hypoxia and reached the peak at 6 h ( P 〈 0.01 ). ② Ginaton inhibited VECs' secretion of ET induced by hypoxia. At the time of 0.5 h after hypoxia, high-concentration Ginaton was more effective than moderate-concentration and low- concentration Ginaton (P 〈0.01 ). At the time of 6 h and 24 h after hypoxia, moderate-concentration Ginaton was more significantly effective than high-concentration and low-concentration Ginaton ( P 〈 0.05 ). CONCLUSION: Ginaton has significant protective effects on hypoxia-induced ET secretion from cultured VECs and the effects of moderate-concentration Ginaton are more obvious compared with those of high-concentration and low- concentration Ginaton.
