摘要
目的:探讨3种溶血磷脂酸(lysophosphatidic acid,LPA)受体(LPA1,LPA2及LPA3)mRNA和蛋白在人卵巢肿瘤组织中的表达状况及意义。方法:采用半定量RT-PCR和Western blot检测LPA1、LPA2和LPA3mRNA以及LPA2和LPA3蛋白在人卵巢上皮性肿瘤组织中的表达水平。结果:RT-PCR结果显示,LPA1mRNA在正常卵巢组织、良性卵巢肿瘤及上皮性卵巢癌中表达阳性率分别为100%、100%、95.7%,差异均无显著性(P>0.05);上皮性卵巢癌组织LPA2mRNA、LPA3mRNA表达阳性率(95.7%;91.3%)显著高于良性卵巢肿瘤(21.3%;21.3%)及正常卵巢组织(15.4%;15.4%)(P<0.01);良性卵巢肿瘤与正常卵巢组织LPA2mRNA、LPA3mRNA表达阳性率差异无显著性(P>0.05);上皮性卵巢癌组织中LPA1mRNA表达率和表达水平高于LPA3mRNA(P<0.05)。Western blot结果显示,上皮性卵巢癌组织中LPA2和LPA3蛋白表达阳性率(95.7%;95.7%)均高于良性卵巢肿瘤(50%;42.9%)和正常卵巢组织表达阳性率(38.5%;38.5%)(P<0.01);良性卵巢肿瘤与正常卵巢组织中LPA2和LPA3蛋白表达阳性率及表达水平相比差异无显著性(P>0.05)。病理资料的相关分析显示,上皮性卵巢癌组织中LPA受体mRNA和蛋白表达与临床病理特征无相关性。结论:LPA1、LPA2和LPA3在卵巢上皮性肿瘤中普遍表达,LPA受体可能参与了卵巢癌的发生发展过程。
Objective: To investigate the lysophosphatidic acid (LPA) receptors expression and their biological significance in human epithelial ovarian neoplasms. Methods: The reverse transcription polymerase chain reation (RT PCR) and Western blot were employed to measure the expression of LPA-1,LPA-2,and LPA 3 receptor mRNA and LPA-2 and LPA-3 receptor protein expression in human epithelial ovarian neoplasms. Results: 1)The positive rate of LPA-1 receptor mRNA in normal ovarian tissue, ovarian benign disease, and ovarian cancer was 100 %, 100 %, and 95.7 %, respectively. The difference was not significant between them(P〉0.05). The positive rates of LPA-2 and LPA 3 receptor mRNA (95.7 % and 91.3 %) were significantly higher in epithelial ovarian neoplasms than in ovarian benign disease (21.3% and 21. 3%) and normal ovarian tissue (15. 4% and 15.4%). The positive rates of LPA-2 and LPA-3 receptor mRNA had no significant difference between ovarian benign disease and normal ovarian tissue(P;〉0.05). The mRNA expression of LPA-1 receptor was markedly higher than LPA 3 receptor in epithelial ovarian neoplasms(P〈0.05). 2)Western blot revealed that the positive rates and expression level of LPA. 2 and LPA-3 receptor protein were significantly increased in epithelial ovarian neoplasms compared with ovarian benign disease and normal ovarian tissue (95.7% and 95.7% vs 38.5% and 38.5% ,P〈0.01). The positive rates and expression level of LPA-2 and LPA- 3 receptor protein had no significant difference between ovarian benign disease and normal ovarian tissue(P〉0.05). 3)No significant correlation was found between LPA receptor expression and clinicopathological features. Conclusion: LPA1, LPA2, and LPA-3 receptors were highly expressed in human epithelial ovarian neoplasms. LPA receptors may be involved in the development and progression of human ovarian cancer.
