芪参降脂饮对动脉粥样硬化大鼠的干预 被引量:2

摘要 目的:研究芪参降脂饮对动脉粥样硬化(AS)的干预,并进一步探讨其可能的机制。方法:24只雄性SD大鼠随机分为3组:阴性对照组、模型组、治疗组,每组各8只,模型组采用高脂饮食配合注射维生素D2建立大鼠AS模型,治疗组在给予模型组相同处理的同时,将芪参降脂饮水煎剂浓缩成1kg/L,按1.5ml/100g体重灌胃,喂饲6周。实验结束,检测各组大鼠血脂和血清丙二醛(MDA)、超氧化物歧化酶(SOD)含量,观察主动脉组织结构变化,测定主动脉壁基质金属蛋白酶-2(MMP-2)的表达及活性。结果:与模型组相比,治疗组大鼠血清的胆固醇、低密度脂蛋白胆固醇及MDA等均显著下降(均P<0.01),SOD含量升高(P<0·01);治疗组大鼠主动脉AS程度明显减轻,主动脉壁MMP-2的表达及活力较模型组降低(P<0·01)。结论:芪参降脂饮干预AS的形成与降脂、抗氧化、抑制MMP-2的表达及活力有关,并推测其具有维持斑块稳定性的作用。 Objective: To study the effect of Chinese medicine "Qishenjiangzhi decoction" (QSJZD) on athero sclerosis (AS) and to elucidate its mechanism further. Method: Twenty-four healthy male SD rats were randomly divided into 3 groups: control group, model group, Chinese medicine treated group. The model rats of atherosclerosis were established by feeding with high cholesterol diet and injecting with vitamin D2 for 6 weeks. Six weeks later, serum lipid levels of MDA, SOD and lipid were measured for all the rats. HE staining was used to study the pathology change of atherosclerosis. Immunohistochemistry and gelatin zymography were used to detect the expression and activity of MMP-2 in aorta vessel. Result: Comparing the chinese medicine treated group with model group, the serum level of TC, LDL-C and MDA were significantly reduced (P 〈 0.01), while serum SOD level was obviously increased (P 〈 0.01). The damage of the aortas in medicine treated group was less severe than that in model group. The expression and activity level of MMP-2 in medicine treated groups was significantly higher than the model groups. (P 〈 0.01). Conclusion: QSJZD could intervene the formation of atherosclerosis. The mechanism might be through suppressing TC, LDL-C and lipid peroxidation, and affecting the activity and expression of MMP-2.
