摘要
目的 建立高效液相色谱法测定人血浆中全反式维甲酸浓度。方法 色谱柱为Supelcosil LC-18DB柱(5μm,4.6mm×250mm);流动相为甲醇-水-冰乙酸(体积比82:17:1);柱温为室温;流速1.5mL·min^-1;检测波长为340nm。结果 全反式维甲酸血药质量浓度在10—1000ng·mL^-1范围内,与峰面积比有良好的线性关系(r=0.9997),最低检测浓度为2.5ng·mL^-1。平均方法回收率为(94.5±4.7)%(n=15)。日内RSD≤4.1%,日间RSD≤6.2%。结论 本方法简单、快速、灵敏、重现性好,适用于全反式维甲酸临床血药浓度监测厦人体药代动力学研究。
Objective To establish a HPLC method for determination of all-trans retinoic acid in human plasma. Methods A Supelcosil LC-18DB column (5μm,4.6 mm × 250 mm) was adopted. Mobile phase was a mixture of methanol,water and glacial acetic acid( 82: 17:1 ). The flow rate was 1.5 mL · min ^-1. The UV detection wave was 340 nm. ResultsThe assay procedure was shown to produce linear calibration curves over the range of 10 ng · mL^-1 to 1000 ng· mL^-1 of all-trans retinoic acid in plasma (r =0.9997). Within the range,the recovery rate was (94.5 ±4.7)% ,and the intra-day RSD and inter-day was less than 4.1% and 6.2% ,respectively. The detection limit of all- trams retinoic acid was 2.5 ng · mL^-1. Conclusions This method is repeatable, convenient and sensitive for plasma concentration monitoring and clinical pharmacokinetics study of all-trans retinoic acid.
