摘要
目的考察透皮吸收促进剂对甲氧沙林脂质体(LMOP)凝胶体外透皮作用的影响。方法分别制备无透皮促进剂的LMOP凝胶及以1%(ω)氮酮和1%(ω)丙二醇为透皮吸收促进剂的LMOP凝胶。采用Franz扩散池,以离体小鼠皮肤为透皮屏障,用高效液相色谱法测定体外接受液中甲氧沙林(8-MOP)的含量,计算其累积透皮释药量(Q),并将数据进行释放动力学模型拟合。结果样品凝胶体外释药符合H iguchi方程,不含透皮吸收促进剂的LMOP凝胶:Q=79.65t1/2+27.99;含透皮吸收促进剂的LMOP凝胶:Q=117.39t1/2+34.37,显示后者的渗透作用更好。结论以1%(ω)氮酮和1%(ω)丙二醇为透皮吸收促进剂的脂质体凝胶的透皮渗透效果更佳。
Objective To evaluate the effects of skin permeation enhancers, 1% (ω ) azone and 1% ( ω ) propylene glycol, on transcutaneous absorption of 8-methoxypsoralen-liposome gel. Methods Gels of 8-methoxypsoralen-liposome with and without skin permeation enhancers were prepared. A piece of excised abdominal hairless mouse skin was set in a Franz-type diffusion cell. Penetration of 8-methoxypsoralen was detected by HPLC and cumulative permeation (Q) calculated and simulated in pharmacokinetics model. Results The drug release and permeation studies in vitro in Franz diffusion cell followed the Higuchi equation with optimum correlation coefficient. For the gel without enhancers, equation was as following: Q = 79. 65t1/2 + 27.99; for the gel with enhancers, equation was as following: Q = 117.39t1/2 + 34.37. Conclusions 8-methoxypsoralen-liposome gel with 1% (ω) azone and 1% (ω) propylene glycol has a higher permeation rate than the gel without enhancers.
