survivin在非小细胞肺癌中表达的意义及其与caspase-3、DFF45蛋白之间的相互关系 被引量:3

摘要 目的探讨survivin蛋白的表达在非小细胞肺癌(NSCLC)发生、发展方面的作用及其与临床病理特征之间的联系,与caspase3及DFF45表达的相互关系。方法运用免疫组织化学SP法对60例NSCLC、10例淋巴结转移癌、10例正常支气管粘膜上皮石蜡切片组织中survivin、caspase3及DFF45蛋白的表达进行检测。结果NSCLC、淋巴结转移癌中survivin的阳性表达率分别为70%及90%,显著高于正常支气管粘膜上皮的阳性率10%(P<0.01)。survivin在NSCLC中的表达与分化程度(P<0.01)、TNM分期(P<0.05)、淋巴结转移(P<0.05)有关。而与年龄、性别、病理类型无明显相关(P>0.05)。survivin表达与caspase3的表达呈负相关(P<0.01;r=-0.335),而与DFF45的表达却呈正相关(P<0.01;r=0.406)。结论survivin在NSCLC的发生、发展中起重要作用,可作为NSCLC的诊断指标,对NSCLC恶性程度的判断及预后也有一定参考价值。survivin的作用是通过抑制caspase3的表达,从而影响下游凋亡途径而发挥作用的。 Objective To investigate the expression of survivin protein in non-small cell lung cancer (NSCLC)and the clinicalpathological characteristic of lung cancer, to evaluate the relationship between expression of survivin, caspase-3 and DFF45 proteins. Methods The expression of surviving and caspase-3, and DFF45 proteins was evaluated by immunohistochemistry in 10 normal bronchial epitheli- um, and 60 NSCLC and 10 metastatic carcinoma of lymph node. Results The expression of survivin protein was detected in a significantly greater proportion in NSCLC(70%) and metastatic carcinoma of lymph node(90%) than normal bronchial epithelium(10%). The up-regulation expression of survivin was related to the TNM stages(P〈0. 05), cell differentiation(P〈0. 01) and lymph node metastases(P 〈0.05), but no relation with age, sex and histological subtype(P〉0. 05). Expression of caspase-3 was negatively(P〈0. 01 ; r = - 0. 335) and DFF45 positively correlated with expression of survivin(P〈0. 01 r = 0. 406). Conclusion survivin may play an important role in the progression of NSCLC and be identified as a defining diagnostic marker, survivin is worthy of analyzing malignant grade and judging the prognosis of NSCLC. Survivin can resist cell apoptosis by restrainting caspase-3 expression.
