EAggEC中小肠结肠炎耶尔森菌强毒力岛结构和功能研究 被引量:1

Investigation of the structure and function of the “high pathogenicity island” of Yersinia enterocolitica WA in enteroaggregative E.coli strains

摘要 目的了解小肠结肠炎耶尔森菌(Yersinia enterocolitica,Yen)强毒力岛在肠聚集性大肠杆菌(EAggEC)中的结构和摄铁功能,为进一步研究细菌毒力的进化奠定理论基础。方法PCR扩增、原位杂交及SDS-PAGE。结果在检测的6株EAggEC有5株检出了毒力岛,而且这些阳性菌株中的毒力岛都位于asntRNA(天门冬氨酸tRNA)位点;在缺铁条件下,EAggEC能够表达和耶尔森菌相同的摄铁蛋白HMWP1和HMWP2。结论小肠结肠炎耶尔森菌强毒力岛在肠聚集性大肠杆菌中有阳性率较高的分布,并具有相同的摄铁功能,很可能EAggEC和耶尔森菌中的强毒力岛有相同的转移机制。 Objective To detect the "high pathogenicity island"(HPI) of Yersinia enterocolitica(Yen) WA in enteroaggregative E. coli, as the theoretical base of bacterial evolution process and the different structure of E.coli. Methods Polymease chain reaction(PCR), nucleic acid hybridization in situ, DNA sequencing and SDS-PAGE. Results Five strains were isolated from 6 EAggEC strains, in these isolates, the HPI bordered by an asntRNA locus, as in Yersinia spp, at the condition of iron deficiency, EAggEC expressed two high molecular weight proteins, HMWP1 and HMWP2. Conclusion The HPI of Yen was found in EAggEC, and the HPI which habored the asntRNA locus has full structure and same iron uptake function as Yersinia, maybe the HPI of EAggEC and Yen shares a same mechanism of transfer.
