摘要
目的对布鲁杆菌OMP28基因进行多态性分析,评价其作为基因标识的价值。方法根据聚合酶链反应-单链构象多态性(PCR-SSCP)分析技术原理,对中国120株布鲁杆菌的OMP28基因进行PCR扩增、酶切、电泳分析和筛选,取不同带型菌株的目的基因进行测序,将结果与Genbank中的布鲁杆菌OMP28序列进行比较。结果 120株布鲁杆菌呈现4种不同图谱(Ⅰ、Ⅱ、Ⅲ、Ⅳ),仅有3个碱基变异,即Ⅳ型与其他3个型第 304碱基A-G,Ⅱ型与另外3个型第405碱基C-T,Ⅰ与其他3个型第498碱基C-T变异,而且均未改变氨基酸序列。结论布鲁杆菌OMP28基因高度保守,具稳定的抗原特征。
Objective To evaluate the value of OMP28 being a gene marker through analyzing the gene type of OMP28 isolated from 120 strains of Brucella. Methods The sequence of OMP28 isolated from 120 strains of Brucella was analyzed using a PCR-SSCP strategy and the results were compared with that of Genbank. Results Four polymorphisms were identified (Ⅰ, Ⅱ, Ⅲ, Ⅳ) in all species examined. Mutations occurred only at 3 bases, which were A-G at the 304th base in type Ⅳ and C-T at the 405th base in typeⅡ and the 498th base in type Ⅰ. The sequence of amino acid kept unchanged. Conclusions OMP2g is highly homological and characterized by stable antigen.
