摘要
以SD大鼠睾丸为实验材料,应用免疫组织化学方法进行染色,探讨免疫组化石蜡切片实验中各条件对实验结果的影响．结果表明,玻片处理、试剂纯净度、取材到封片等,都对实验结果会产生较大影响．而脱水、抗原修复、过氧化氢、一抗及正常羊血清孵育过程是影响实验的关键因素．
In this study, the SD mouse testis and immunohistochemical methods were used to study the effects of different experimental conditions on the result. The results indicated that the treatment of the glass slides, the purity degree of the reagents, and the procedure from obtaining specimen to sealing the tissue sections, during these processes, every step affect the experimental result, but we found the following processes are the critical influences to the experimental result, which include the procedure of desiccation, antigen retrieval, Hydrogen peroxide incubation, primary antibody incubation, and normal goat blood serum incubation.
