摘要
目的:阐明心脏肾素-血管紧张素系统(RAS)在左室肥厚形成中的重要作用,探讨转换酶抑制剂──依那普利逆转左室肥厚的机制。方法:将雄性15周龄自发性高血压大鼠(SHR)随机分为治疗组(n=7)和对照组(n=6),分别以管饲法给依那普利(溶解在蒸馏水中,其浓度为10mg/ml)20mg·kg-1/日和等量的蒸馏水治疗5周。以放射免疫法和高效液相层析法测定血管紧张素Ⅱ(AngⅡ)和去甲肾上腺素(NE)浓度。以尾套法测血压。结果:治疗组左室肥厚逆转,血压下降,同时伴左室心肌组织AngⅡ含量降低,血浆AngⅡ水平升高。左室心肌NE总含量和前肢肌肉组织中NE浓度无明显改变。左室+室间隔重/体重与左室心肌组织AngⅡ含量呈显著正相关(r=0.626,P<0.05)。结论:心脏RAS在左室肥厚形成过程中起着重要作用,心肌组织NE可能不参与依那普利逆转心肌肥厚的机制。
Objective:To study the roles of the cardiac renin-angiotensin system (RAS) and converting enzyme inhibitor-enalapril in left ventricular hypertrophy (LVH).Methods:Enalapril(20 mg/kg per day)in distilled water 10 mg/ml and equal volume of distilled water were administered to 15-week-old male spontaneously hypertensive rats (SHR) of the experimental (n=7)and control (n=6) groups respectively for 5 weeks by gavage. Blood pressure,plasma and ventricular angiotensin Ⅱ(Ang Ⅱ) and tissue norepinephrine (NE) were measured by tail-cuff method,radioimmunoassay,and high-performance liquid chromatography,respectively.Results:Long term treatment with enalapril in SHR decreased the blood pressure and Ang Ⅱ level of the left ventricle(p<0.01)which was associated with the regression of LVH,but increased the plasma Ang Ⅱ level (p<0.01).There was a significant positive correlation between the left ventricular Ang Ⅱ level and the ratio of left ventricular weight to body weight (r=0.626,p<0.05).While the left ventricular NE content and forelimb muscle NE concentration showed no significant difference between the two groups.Conclusions:Cardiac but not plasma Ang Ⅱ level may be an important factor in the pathogenesis of LVH in SHR. Cardiac tissue NE is not influenced by enalapril and is probably not involved in the regression of LVH.
