摘要
为探讨人工合成的三肽囊素(BS)作为新型免疫增强剂的作用机制,本研究采用MTT比色法、放射免疫测定法(RIA)和相对半定量RTPCR法,分别研究了BS对体外培养的鸡脾脏淋巴细胞的增殖、对白细胞介素2(IL2)和白细胞介素6(IL6)分泌及其mRNA表达水平的影响。试验结果表明,0.5μg/ml、5μg/ml和50μg/mlBS处理36h,均可显著提高ConA诱导的脾脏淋巴细胞的增殖反应性;5μg/mlBS处理能显著促进脾脏淋巴细胞分泌IL6,显著升高IL6mRNA表达水平,但对IL2的分泌及其mRNA表达水平无显著影响。本研究结果进一步提示,BS可能主要通过激活IL6基因的表达和促进脾脏淋巴细胞的增殖反应。
Immunostimulatory effects of synthetic bursin (BS) on proliferation of chicken splenic lymphocytes and gene expression of interleukin-2 (IL-2) and interleukin 6 (IL-6) were studied by MTT colorimetry, RIA and semi-quantitative RT-PCR. The results showed that treatment with 0.5μg/ml, 5μg/ml or 50μg/ml of BS for 36 h and then with 10μg/ ml of Concanavalin A (Con A) for 24 h promoted significantly proliferation of chicken splenic lymphocytes. The concentration of IL-6 in the cell supernatant of BS-treated cells was significantly higher than that of control group, while the concentration of IL-2 had no significant difference following treatment with 5μg/ml of BS. Under the same conditions, simi- lar results were obtained by semi-quantitative RT-PCR for mRNA expression levels of IL-2 and IL-6. These data suggest that bursin exerts its stimulatory effect on humoral immune response mainly by promoting expression of IL-6 and proliferation of chicken splenic lymphocytes [Acta Zoologica Sinica 52 (1): 170 - 174, 2006].
