重组人γ─干扰素联合肿瘤坏死因子或高热体外净化实验研究 被引量:3

Experimental study on purging in vitro with recombinant human interferon gamma and tumour necrosis factor or hyperthermia

摘要 用重组人γ-干扰素(IFN)和肿瘤坏死因子(TNF)体外净化骨髓中残存的白血病细胞时,1000U/mlIFN或100U/mlTNF单独应用时仅能清除大约80%左右,而二者联用时则能起到协同抗白血病的效应,可清除90%以上残存的白血病细胞,但对CFU-GM的形成却没有协同抑制作用,抑制率仅为54%。在高热体外净化研究中,单独加热到41℃作用2h,清除率达90%,若同时加入1000U/ml的IFN,可大大增强高热的净化效果,清除率可达98%以上,且减轻了高热对CFU-GM的抑制作用,CFU-GM的抑制率下降为24.5%,与单纯高热净化时的32.8%相比,P<0.05。 When bone marrow was purged in vitro with recombinant human interferon gamma (rhIFN γ ) 1000 U/ml or tumour necrosis factor(TNF)100 U/ml alone,about 80% of the residual leukemic cells in it could be eliminated.Combination of rhIFN γ and TNF showed a synergic elimination of the residual leukemic cells with a clearance rate of above 90%,but no synergic inhibition of the normal bone marrow CFU GM(inhibitory rate was only 54%)was observed.Hyperthermia(41℃)alone could eliminate more than 90% residual leukemic cells,but when IFN 1000 U/ml was added,the elimination rate was raised to above 98%,while the inhibitory rate of the growth of normal bone marrow progenitor(CFU GM)was decreased to only 24.5% which was much lower than that (32.8%)caused by hyperthermia(41℃),so it is better to raise the temperature for purging in vitro to remove the residual leukemic cells from bone marrow.
