摘要
目的探讨小檗碱降低2型糖尿病大鼠血清β样淀粉蛋白(Aβ)的作用机制,为防治2型糖尿病并发阿尔茨海默病(A lzhe im er’s d isease)的研究提供新的思路。方法用链脲佐菌素静脉注射+高脂、高热饲料喂养2周的方法复制2型糖尿病大鼠模型20只,随机分为2型糖尿病组和小檗碱治疗组(盐酸小檗碱剂量:187.5 mg.kg-1.d-1)各10只;另设正常对照组大鼠10只。2型糖尿病组和小檗碱治疗组继续用高脂高糖饮食饲养6周后处死,分离血清及脑组织,分别测定Aβ、乙酰胆碱酯酶(AchE)、丁酰胆碱酯酶(BchE)活性、空腹血糖(FBG)、空腹胰岛素(FINS)水平。结果2型糖尿病组血清AchE、BchE、Aβ、FBG、FINS均显著高于正常对照组,脑组织Aβ也呈升高趋势,但AchE、BchE活性明显下降;小檗碱治疗组血清AchE、FBG、Aβ显著下降,FINS水平也明显降低。结论2型糖尿病大鼠血清AchE、BchE活性显著增高,Aβ含量明显升高,提示2型糖尿病与阿尔茨海默病存在着共同的分子基础,二者之间有密切的联系。小檗碱能降低FBG、Aβ水平,可能通过抑制血清胆碱酯酶的活性,改善胰岛素抵抗,因此,对防治2型糖尿病并发阿尔茨海默病具有重要的临床价值。
Objective To probe into the effect of berberine on β-amyloid protein(β-AP) in rats with type-2 DM and its mechanism in order to provide a new idea to prevent and treat type-2 DM complicated by Alzbeimer disease(AD). Methods Twenty type-2 diabetic rat models were duplicated by high-fat, high - calories diet and a very small dose of streptozotocin (STZ) for 2 weeks, and randomly divided into type-2 DM group and berberine treatment (187.5 mg·kg^-1·d^-1) group, 10 rats in each group. A control group of 10 normal rats was also established. After continued feeding for 6 weeks, serum and brain tissue of rats were separated, with their levels of β-AP, AchE, BchE, FBG and FiNS determined. Results The serum levels of AchE, BchE, β-AP, FBG, FINS in DM group were significantly higher than those of the control group. The level of β-AP in the brain tissue was increased and the activities of AchE and BchE were decreased. The levels of AchE, FBG, β-AP and FINS were decreased in treatment group with berberine. Conclusion Rats with type-2 DM has a tendency of over-expression of AchE, BchE and β-AP. suggesting that type-2 DM may share a common pathological mechanism with AD. Berberine can lower the level of FBG and β-AP, probably via its inhibition on the activity of serum chonlinesterase, thus improving insulin resistance. Therefore, berberine is important clinically in the preveation and treatment of type-2 DM complicated by AD.
