空间微载体细胞培养技术的地面研究

摘要 为建立适合空间应用的细胞培养技术,利用自行研制的细胞培养反应器,采用微载体悬浮培养系统,对L929细胞和CHO细胞进行了培养。结果表明:在面积相同条件下,微载体悬浮培养的细胞产量是不加微载体单层培养的26倍;微载体浓度为2m/ml,相对高一些细胞接种密度;10%左右的血清含量培养基为较优的细胞培养条件。为适合空间条件的需要,采用倍增补液法与微载体球间细胞转移法,探索了细胞扩增培养的途径。同时,对适合空间细胞培养的反应器进行了探索研究。 Two cell lines L929 and CHO were cultured in cell bioreactor with microcarrier suspension culture system. The optimum conditions for cell proliferation were studied.The result showed that microcarrier density 2 mg/ml,an even higher cell moculation density,sernm density abeut 10% were suitable for cells to grow. The cell production under such conditions with microcarrier suspension techniques were 26 times more than that with mono-layer culture. Scale-up of culture volume and beads to beads transfer techniques were devel-oped and new bioreactor were explored to be used in space.
