摘要
目的:通过观察杭白芷香豆素(CAD)对戊巴比妥钠和巴比妥钠催眠作用的影响,初步探讨其中枢作用及其发生机制。方法:将小鼠随机分为溶媒对照组(5%吐温-80;20 ml/kg)、氯霉素阳性对照组(25 mg/kg)和CAD组(25,50,100 mg/kg)。等容量灌胃给药1 h后,再腹腔注射戊巴比妥钠40 mg/kg或巴比妥钠200 mg/kg,观察小鼠催眠潜伏期和睡眠时间。结果:氯霉素组和3个剂量的CAD组均可使戊巴比妥钠睡眠时间延长,分别延至溶媒对照组的380%、284%、353%和366%(P<0.01);100 mg/kg CAD组还可显著延长巴比妥钠催眠潜伏期(P<0.01),并缩短其睡眠时间(P<0.01)。结论:CAD具有一定中枢兴奋作用,但同时可能具有肝药酶抑制作用。
Objective: To survey the influence and mechanism of the coumarins from Radix Angelicae Dahuricae (CAD) on the hypnotic effects of pentobarbital sodium and barbital sodium in mice. Methods: Sixty Kunming mice were randomly divided into five groups, with 12 animals in each group. Mice in group 1, 2, 3, 4 and 5 were given each with 20 ml/kg gastrogavage (5% tween80), 25 mg/kg of chloromycin, and 25,50,100 mg/kg of CAD in equal volumes, respectively. One hour later, all of the animals were given'each an intraperitoneal injection of 40 mg/kg of pentobarbital sodium or 200 mg/kg of barbital sodium. The latent period and duration of hypnosis of the mice were recorded under the observation. Results: The duration of hypnosis induced by pentobarbital sodium was prolonged by 380%, 284%, 353% and 366% (P〈0.01), respectively, in mice given chloromycin and three different doses of CAD. 100 mg/kg of CAD was shown to significantly prolonged the latent period (P〈0.01) and shortened the duration of hypnosis (P〈 0.01) induced by barbital sodium. Conclusion: CAD could inhibit the liver microsomal cytochrome P450 activity. CAD in the higher dose seems to excite the central nervous system in mice.
