摘要
腺苷酸活化蛋白激酶(AMPK)作为机体能量监测器,其活性主要受AMP/ATP比例调控。通过抑制能量合成,加速能量分解和ATP合成以保持机体能量平衡的同时,AMPK同样参与了胰岛β细胞胰岛素分泌的调节,并直接作用于β细胞的相关基因表达和胰岛素信号转导。以二甲双胍、罗格列酮、瘦素和脂联素为代表的AMPK激活剂对胰岛素分泌的直接影响尚有争议。本文就AMPK介导的胰岛β细胞功能作一简述。
As a key sensor of cellular energy status in vivo, the action of 5'-AMP-activated protein kinase (AMPK) is mainly regulated by the AMP/ATP ratio. AMPK can maintain the energy balance in body through inhibiting anabolism, accelerating catabolism and synthsis of ATP. Furthermore, AMPK participates in regulating the secretion of insulin from pancreatic β cells, and affects the gene expression and insulin signaling transduction as well. Agents which can activate AMPK, including metformin, the glitazones, leptin and adiponectin may be expected to affect the insulin secretion directly. However, the conclusion is still disputed. This article reviews the recent studies focusing on the relationship between AMPK and the pancreatic β cell function.
