Insulin signal transduction pathway in pancreatic β-cell

摘要 胰岛β细胞上存在着胰岛素受体(IR)及胰岛素受体底物(IRS),这些蛋白及其下游信号蛋白构成了复杂的信号转导通路,调控胰岛素的分泌,维持β细胞的生长、增殖和存活。其主要途径是磷脂酰肌醇3激酶(PI3K)信号转导途径和钙通道及其相关途径。β细胞IR、IRS基因敲除鼠表现出糖耐量异常或糖尿病症状。对这一信号通路的深入研究有利于进一步阐明糖尿病的发病机制并为糖尿病的治疗提供新思路。 Insulin receptor (IR) and insulin receptor substrate (IRS) are diseoveried in pancreatic β-cell. These proteins and their downstream signal proteins construct a complex signal transduction pathway, which regulates the secretion of insulin, maintains the growth, proliferation and survival of β-cell. The major pathways are phosphatidylinositol 3-kinase pathway, Ca^2+ channel and its related pathway. IR/IRS gene in β cell knock-out mice show impaired glucose tolerance or diabetes. Further study of these pathways will be useful to elucidate the pathogenesis of diabetes and provide a new idea in the treatment of diabetes.
