神经肽P物质促进增生性瘢痕中肥大细胞组胺释放的定量检测 被引量:2

摘要 目的:神经肽P物质可以促进人增生性瘢痕中肥大细胞组胺的释放,定量分析在此过程中神经肽P物质浓度因素的作用,探讨增生性瘢痕中神经肽P物质与肥大细胞的相互作用及作用条件。方法:实验于2005-01/10在解放军第三军医大学西南医院中心实验室进行。将取自于西南医院整形美容外科烧伤患者的增生性瘢痕活体组织块切下后立即处理(患者或监护人同意),修剪成0.5mm×0.5mm×0.5mm~1mm×1mm×1mm大小,将组织块放入0.5mL含10-6,5×10-6,10-5,5×10-5,10-4mol/L神经肽P物质的DMEM液中,以0mol/L神经肽P物质为对照组。组织块作用30min后提取上清液,为样品组胺;提取上清液后的组织块分别加入去离子水0.5mL,加热煮沸(破坏细胞,释出组胺)20min,摇匀后离心(1500g,10min),吸出上清液,为剩余组胺。荧光法测定作用后上清液中肥大细胞的组胺释放率[组胺释放率=样品组胺/(样品组胺+剩余组胺)×100%]。结果:神经肽P物质10-6,5×10-6,10-5,5×10-5,10-4mol/L组组胺释放率显著高于对照组[(49.78±3.56)%,(54.56±1.90)%,(57.12±1.49)%,(60.49±1.41)%,(63.16±2.61)%,(43.96±3.18)%,P<0.01],且神经肽P物质浓度越高,组胺释放率越高(P<0.01或0.05)。结论:在增生性瘢痕中,神经肽P物质以剂量依赖方式刺激肥大细胞组胺的释放,当神经肽P物质达到10-6mol/L时即有肥大细胞组胺的显著释放,随神经肽P物质浓度提高,组胺释放率升高。 AIM: Neuropeptide substance P can accelerate the release of mastocyte histamine in human hyperplastic scar, To analyze quantitatively the effect of concentration of neuropeptide substance P in this process and explore the interaction and effect conditions between substance P and mastocytes in hyperplastie scar. METHODS: The experiment was conducted from January to October in 2005 at central laboratory, Southwest Hospital, Third Military Medical University of Chinese PLA. Hyperplastie scar tissues of burn patients from Plastic Surgery, Southwest Hospital were cut into 0.5 mm×0.5 mm×0.5 mm to 1 mm×l mm×1 mm size. Patients or their guardian agreed to take part in the experiment. The tissue block was put into 0.5 mL DMEM liquor with 10^-6,5×10^-6, 10^-5,5×10^-5 and 10^-4 mol/L substance P. 0 mol/L substance P was considered as control group. After 30 minutes, supernatant was extracted as sample histamine, and then tissue blocks were added with 0.5 mL soft water, heated, and boiled for 20 minutes (destroying the cells and set free histamine). After shaking evenly, centrifuged (1 500 g for 10 minutes). Supernatant liquor was sucked, that was, the leftover histamine. The release rate of histamine of mastocytes in supernatant was detected with fluorescent method [release rate of histamine=sample histamine/(sampie histamine ± leftover histamine)×100%]. RESULTS: The release rate of histamine in the substance P 10^-6,5×10^-6, 5×10^-5, 10^-4 mol/L group was higher markedly than that in the control group [(49.78±3.56)%, (54.56±1.90 )%, (57.12± 1.49 )%, (60.49±1.41 )%, (63.16±2.61 )%, (43.96±3.18)%,P 〈 0.011. The higher the concentration of substance P, the higher the release rate of histamine was (P 〈 0.01 or 0.05). CONCLUSION: In hyperplastic scar, the dose-dependent substance P can stimulate the release of histamine in mastocytes. When the substance P reaches 10^-6 mol/L, there are a lot of significant releases of histamine in mastocytes. With the increase of the concentration of substance P, the release rate of histamine elevates, also.
