摘要
目的:分析中国北京社区人群脂联素基因+45T/G和过氧化物酶增殖物活化受体γ2基因Pro12Ala多态性及其交互作用与2型糖尿病的关系。方法:自2001年解放军总医院老年医学研究所“对万寿路等社区的流行病学调查”人群中选出无亲缘关系2型糖尿病患者(糖尿病组)195例,非糖尿病对照(非糖尿病组)139例。应用聚合酶链反应-限制性片段长度多态性方法分析两组受试者脂联素+45T/G与过氧化物酶增殖物活化受体γ2Pro12Ala多态性各基因型分布频率,用Logistic回归分析了解基因间的交互作用。结果:334例全部进入结果分析。①脂联素+45T/G多态性各基因型分布频率为:糖尿病组,TT52.8%,TG35.4%,GG11.8%;非糖尿病组,TT56.1%,TG41.0%,GG2.9%,糖尿病组GG型明显多于非糖尿病组(P<0.01)。②过氧化物酶增殖物活化受体γ2Pro12Ala多态性各基因型分布频率为:糖尿病组PP90.8%,PA9.2%;非糖尿病组PP89.9%,PA9.4%,AA0.7%,两组间各基因型频率分布差异无显著性(P>0.05)。③两位点交互作用组间比较,具有脂联素+45TG+GG型和过氧化物酶增殖物活化受体γ2PA+AA型者患2型糖尿病的风险较高(P<0.05)。结论:脂联素+45T/G多态性与2型糖尿病存在相关性,其纯和突变型GG型罹患率较高;未发现过氧化物酶增殖物活化受体γ2Pro12Ala多态性与2型糖尿病有关;上述两位点存在交互作用,携带脂联素+45G等位基因和过氧化物酶增殖物活化受体γ2Ala12等位基因者易患2型糖尿病。
AIM: To analyze the interaction of adiponectin +45T/G and peroxisome proliferator-activated receptor (PPAR) γ2 Prol2Ala polymorphisms and their relationship with type 2 diabetes in population of Beijing community of China.
METHODS: 195 type 2 diabetic subjects (diabetic group) and 139 nondiabetic subjects (non-diabetic group) without kinship were selected from Institute of Geriatric Medicine, General Hospital of Chinese PLA since 2001, who were investigated on epidemiological study in the Wanshousi etc, communities. Distribution frequencies of every genotype of adiponectin +45T/G and PPAR γ2 Pro12Ala polymorphisms of the testees of the two groups were analyzed with polymerase chain reaction-restriction fragment length polymorphism, Interaction among genes was analyzed and known with Logistic regression analysis.
RESULTS: 334 patients were all involved in the result analysis, ① Distribution frequencies of every genotype of adiponectin +45T/G polymorphisms: In the diabetic group it was TT 52,8% ,TG 35.4% and GG11.8%; In the non-diabetic group, it was TT 56.1%,TG 41.0% and GG 2.9%, and the GG type in the diabetic group was more significantly than that in the non-diabetic group (P 〈 0.01). ②Distribution frequencies of every genotype of PPAR γ2 Pro12Ala polymorphism: In the diabetic group, it was PP 90.8% and PA 9.2%; In the non-diabetic group, it was PP 89.9%,PA 9.4% and AA 0.7%, and there was insignificant difference of distribution of genetype in the two groups (P 〉 0.05), ③After the comparison of interaction of the two sites, the risk of attack type 2 diabetes in the people with adiponectin +45 TG+GG and PPAR γ2 PA+AA was high (P 〈 0.05).
CONCLUSION: The adiponectin +45T/G polymorphism is associated with type 2 diabetes, and subjects with homozygous mutant GG genotype have high attack rate. Lack of evidence shows that PPARγ2 Pro12Ala genetic polymorphism is associated with type 2 diabetes. The abovementioned sites have interaction. Subjects with adiponectin +45 G allele and PPARγ2 Ala12 allele have higher risk of attacking type 2 diabetes.
