摘要
目的 探讨降钙素基因相关肽(CGRP)对系统性红斑狼疮(SLE)患者外周血单一核细胞(PBMC)凋亡的影响。方法 应用流式细胞仪膜联蛋白(Annexin)V/碘化丙啶(propidineiodide,PI)染色法检测SLE患者、健康献血员PBMC与不同浓度的CGRP和(或)CGRP的受体拮抗剂CGRP8-37培养48h的凋亡百分率。结果 SLE患者PBMC凋亡率显著高于健康献血员(P〈0.001).不同浓度(0.5×10^-8~1.5×10^-8moL/L)的CGRP对健康献血员PBMC凋亡无影响(P〉0.05);但可以不同程度的抑制SLE患者PBMC的凋亡(P〈0.05~0.001);当CGRP浓度超过1.0×10^-8moL/L时,其对SLE患者PBMC凋亡的抑制效应进入平台,增加CGRP浓度并不能进一步显著抑制SLE患者PBMC地凋亡。无论何种浓度的CGRP均不能使SLE患者PBMC的凋亡率降低至正常范围内。CGRP8-37本身对PBMC凋亡无影响(P〉0.05),但是可以桔抗CGRP对SLE患者PBMC凋亡的影响。结论 CGRP通过受体影响PBMC地凋亡;CGRP浓度地降低在某种程度上可导致SLE患者PBMC地凋亡加快。
Objective To study effects of calcitonin gene related peptide (CGRP) on apoptosis of peripheral blood mononuclear cells (PBMC) from patients with systemic lupus erythematosus (SLE). Methods The percentage of apoptosis of PBMC from SLE patients and healthy blood donors was examined under flow cytometry by means of Annexin-V/propidine iodide (PI) staining 48h after cultured with CGRP and/or its receptor antagonist CGRP8-37 at different concentrations. Results The percentage of apoptosis of PBMC from SLE patients was higher than that of healthy blood donors( P 〈 0. 001 ). CGRP, at all tested c, oncentrations, had no effects on apoptosis of PBMC from healthy blood donors( P 〉 0.05 ) ,but significantly inhibited apoptosis of PBMC from SLE patients beyond the spontaneous level at concentrations of 0.5 × 10^-8 mol/L or higher( P 〈 0.05 - 0. 001 ) ,and the effect attained a plateau at 1.0 × 10^-8 rnol/L,which inhibition could not significantly increase as its concentration increased. However, CGRP, at any concentrations tested,couht not reduce the rate of apoptosis of PBMC from SLE patients to normal range, CGRP8-37 alone had no effects on apoptosis of PBMC( P 〉 0.05 ) , but was found to be able to inhibit the effects of CGRP on PBMC. Conclusion CGRP plays an important role in the apoptosis of PBMC from SLE patients via its receptor;low serum levels of CGRP may cause accelerated apoptosis.
