肌源性干细胞在骨创伤修复中的应用

摘要 目的:肌源性干细胞是近来从成年动物骨骼肌中分离出的一类成体干细胞,它可能参与多种组织的修复过程。推测肌源性干细胞可能参与骨缺损的修复。资料来源:应用计算机检索Medline及ScienceDirect(Elsevier),EBSCOhost(ASP),KluwerOnline等数据库1999-01/2005-01相关肌源性干细胞与骨创伤修复的文章,检索词“muscle-derivedstemcell,bone,bonedefect,fracture,boneregeneration,osteogenesis”。应用计算机检索中国期刊全文数据库2003-01/2005-02的相关肌源性干细胞与骨创伤修复的文章,检索词“肌源性干细胞,骨折,骨形成”。资料选择:对资料进行初审,纳入标准:①肌源性干细胞的起源、分布、分化、参与各种组织修复特别是骨创伤修复的文献。②不排除是否为随机、盲法等论证推荐的文章。排除标准:不采用重复研究、个案报告、综述、Meta分析的文献。资料提炼:共检索到相关文献152篇,分别对肌源性干细胞的起源、分离方法、分化、鉴定、参与各种组织修复的情况进行探讨,选用其中具有代表性的30篇。资料综合:肌源性干细胞是具有多向分化潜能的成年干细胞,存在于哺乳动物的骨骼肌中,它可分化为骨髓造血细胞、软骨细胞、成肌细胞、脂肪细胞、成骨细胞及骨细胞等。结论:肌源性干细胞参与骨创伤修复,可以作为骨折愈合或骨不连以及骨缺损修复的种子细胞。 OBJECTIVE: Muscle-derived stem cells, which are a kind of adult stem cells, are isolated from skeletal muscle of adult mammals in recent years. It probably has a role in the repair of multiple tissues. It is postulated that muscle-derived stem cells probably have a role in the repair of bone defects. DATA SOURCES: Literatures relevant to muscle-derived stem cells and traumatic bone repair in the databases of Medline, Science Direct (Elsevier), EBSCOhost (ASP) and Kluwer Online, etc. from January 1999 to January 2005 were searched by computer, and the keywords used for searching were "muscle-derived stem cell, bone, bone defect, fracture, bone regeneration, osteogenesis". Literatures relevant to muscle-derived stem cells and traumatic bone repair in the China Journal Full-text Database (CJFD) from January 2003 to February 2005 were searched by computer with the keywords "muscle-derived stem cell, fracture, osteogenesis". STUDY SELECTION: Primary selection was performed by following inclusive criteria: ① literatures relevant to the origin, distribution, differentiation of muscle-derived stem cells, and those related to its roles in tissue repair, especially bone trauma repair, and ②literatures on randomized, blinded designation or not were not excluded. Exclusive criteria were: literatures on repeated researches, case reports, reviews or Meta analysis were not included. DATA EXTRACTION: A total of 152 literatures were obtained, in which the origin, methods of isolation, differentiation, characterization of muscle-derived stem cells and its roles in the repair several kinds of tissue were explored, respectively. Thirty representative literatures were selected for the review. DATA SYNTHESIS: Muscle-derived stem cells were pluripotent adult stem cells, which resided in the skeletal muscle of mammals. It could differentiate into hematopoietic cells, chondroetyes, myoblasts, adipoctyes, osteoblasts and osteocytes, etc.CONCLUSION: Muscle-derived stem cells participated in the repair of bone trauma, and can be used as seed cells in the repair of fracture, delayed union, non-union, or bone defects.
