Improvement of ICI174, 564 and Nor-binaltorphimine to HemodynamicsFollowing Hemorrhaqic Shock in Rabbits

摘要 用兔侧脑室给药后观察了 -阿片受体特异性拮抗剂ICI174,864和 阿片受体特异性拮抗剂Nor-Binaltorphimine(Nor-BNI)对失血性休克兔血流动力学指标的影响,旨在初步阐明新型阿片受体特异性拮抗剂是否具有抗休克作用。结果显示,ICI174,864(50 g,icv)和Nor-BNI(50g,icv)均可显著改善失血性休克兔各血流动力学指标,包括血压(MAP),左室内压(LVSP),左室内压最大上升和下降速度(±dp/dtmax),心肌收缩速度(Vce40,Vpm)和心肌收缩向量环面积(Lo)。结果表明,特异性 和 阿片受体拮抗剂有一定的抗休克作用,值得进一步研究。 Abstract The present study was aimed to investigate whether ICI174, 864, a highly selectivedelta( ) opioid receptor antagonist, and Nor-binaltorphimine (Nor BNI ), a highly selective kappa(K) opioid receptor antagonist could reverse shock. Effects of ICIl74, 864 and Nor-BNI on hemodynamics following hemorrhagic shock in rabbits were observed. It was found that. ICI174,864(50ug, icv ) could significantly improve the hemodynamic parameters of shocked rabbits. Thesehemodynamic parameters include mean arterial pressure (MAP), left intera-ventricular systolicpressure (LVSP), maximal rate of the change of left intraventricular pressure ( dp/dtmax),velocity of contiactile element shortening (Vec40, Vpm) and area of P-dp/d. vector loop (L0),and so on. These findings indicate that and opioid receptor antagonists have some beneficaleffects on hemorrhagic shock.
