摘要
对核桃离体培养中外植体的褐变进行了初步研究,结果表明:4月份取嫩枝茎段进行培养,效果最差,褐变死亡率高,取6月份半木质化茎段进行培养,效果较好,休眠芽次之。液体状态的培养基可以减轻外植体褐变。培养基中附加激素6-BA 和 IAA、GA 时,外植体的褐变程度较低,外植体腋芽萌发与生长分化较好。培养基中加入硫代硫酸钠对防止外植体褐变,提高外植体萌芽率和有效新梢率有促进作用。
Study was carried out on the factors resulting in browning of walnut explants in vitro culture. The result showed that new shoots collected in April have severer browning and a higher death rate while the shoots collected in June are more effective for culture, with the dormant buds to be second to them. In vitro culture, the browning of explants could be reduced with the liquid medium. With the help of hormones added at proper concentrations (6-BA 1.0 mg/L,IAA 0.2 mg/L or GA 0.5 mg/L) explants can have a better germination of auxiliary buds, with the browning phenomena lessened. Addition of Na2S2O3 to medium can effectively prevent walnut explants from browning, and promote germination and shoot growth.
