摘要
目的观察胰腺癌细胞放疗抵抗与Survivin表达间的关系。方法胰腺癌细胞室温下经不同剂量的X线辐照处理。实验分为A组:对照组,无照射;B组:2Gy照射;C组:半致死量4Gy照射;D组:致死量10Gy照射。RT-PCR法检测各组Survivin mRNA表达量;MTT法检测胰癌细胞增殖活性及流式细胞术检查细胞凋亡。免疫组化法检测38例胰腺癌患者中Survivin蛋白的表达。结果胰腺癌患者中Survivin蛋白的表达阳性率为79%(30/38)。在X线辐照处理后1~2d,B组与C组的Survivin表达明显增加,且在4d达到高峰;D组的Survivin表达显著下降。X线照射后第6天,B组与C组的胰腺癌细胞的增殖未受抑制,而D组的增殖受到明显抑制。B组的凋亡率为3%;C组的凋亡率为5%;D组的凋亡率为53%,与B组、C组相比,差异有显著性(P<0.01)。结论放疗后诱导的Survivin表达上调可能是胰腺癌细胞对放疗抵抗的重要原因。
【Objective】 To observe the relation between radioresistance of pancreatic cancer(PC) cells and expression level of Survivin. 【Methods】 Cells were X-irradiated with vavarious doses of X-rays. A group: control group, no irradiation; B group: the dose of X-irradiation was 2 Gy; C group: the dose of X-irradiation was 4 Cy (Sublethal closes); D group: the close of X-irradiation was 10 Gy (Lethal doses). Survivn protein was detected by immunohistochemical method. Survivn mRNA were measured by quantitative RT-PCR assays. Proliferation of cells was determined using MTT assay and apoptosis was determined by flowcytometry. 【Results】 The expression of Survivin protein was detected in 30 of 38 patients with PC (79%). The expression level of Survivin mRNA of PC cells gradually increased after 2 days exposure to X-irradiation in groups B and C, peaking at 4 days; but it was obviously decreased in D group, After 6 days exposure to X-irradiation, the proliferation of PC cells was not obviously suppressed in groups B and C, lint was significantly increased in D group. The apoptosis rates was 3% in B group, was 5% in C groups, was 53% in D group. 【Conclusion】 Radiation therapy induced augmentation of Survivin expression maybe was the, important factor causing radioresistance in PC cells.
