摘要
目的探讨低剂量环磷酰胺(CY)对脓毒症大鼠急性肺损伤(ALI)肺组织的影响。方法采用急性肺损伤大鼠模型,将40只SD大鼠随机分为四组:实验组(CY组)、阳性对照组(DXM组)、阴性对照组(ALI组)和空白对照组(NS组),每组10只。建模成功6 h后留取肺组织,用免疫组织化学法结合图象分析仪检测肺组织NF-κB p65蛋白的相对含量,并进行肺组织的病理学光镜检查。结果ALI组病理切片HE染色光镜下见:肺泡腔见大量出血及炎性细胞浸润,支气管上皮剥脱、水肿等。DXM组、CY组也可见炎细胞浸润、肺萎陷现象,与ALI组比较,明显减轻(P<0.05)。CY组、DXM组与ALI组比较,大鼠肺组织NF-κB p65蛋白表达明显降低(P<0.01),IκB-α表达显著升高(P<0.05);CY组和DXM组比较无差异(P>0.05)。结论低剂量CY减轻了ALI大鼠肺组织中性粒细胞的浸润,能明显下调NF-κB p65蛋白表达。低剂量CY与地塞米松一样具有明显的抗炎作用,能够减轻内毒素诱导的ALI大鼠的肺组织损害。
Objective To explore the effects of low dose of cyclophosphamide (CY) on acute lung injury (ALl) in sepsis rats. Methods ALl rat models were given the intrapefitoneal injection of lipopolysaccharide(LPS). 40 SD rats were randomly divided into four groups: experiment group(CY group, ALl + CY, n = 10), positive control group( DXM group, ALl + DXM, n = 10), negative control group (ALl group, ALl + NS, n = 10) and uncontrolled group(NS gwoup, n =- 10). All the rats were killed and the specimens of lung tissues were reserved at 6 hours after establistunent of the models. The relative content of NF- κB P65, IκB- α were detected by image analyse system and immunohistochemistry. The pathological changes of the lurg tissues were exanined with light microscope. Results By light microscopy, the evidence of lung injury - neutrophil infiltration and hemorrhage in the cavity of 'alveolus, bronehiole epithelial desquamation, edema and so on were found in the sections of ALl group stained with hematoxylin- eosin. Compared with ALl group, neutrophil infiltration, alveolar atelectasis in CY group and DXM group, were alleviated( P 〈 0.05 ) ; CY group and DXM group were compared with ALl group, the expression of NF - κB P65 was down - regulated ( P 〈 0.01 ) and IκB - α was up - regulatod ( P 〈 0.05 ) . CY group was compared with DXM group , the difference wasn't found( P 〉 0.05). Conclusions Low dose of CY alleviated neutrophil in filtration in lung, and down - regulated the expression of NF κB P65. The result indicated that low dose ofCY, same as DXM had prominently anti inflammatoly effect and can relieve the hmg injury in ALI rats induced by LPS.
