绿色蔬菜叶绿体DNA的提取和纯化 被引量:6

Extraction and Purification ctDNA in Green Leaves Vegetables

摘要 对蔬菜绿叶通过组织匀浆,差速离心分离,密度梯度离心分离、纯化、提取叶绿体。显微镜下观察所得叶绿体纯度、形态及结构,看到椭圆、球形叶绿体,并用数码照相机拍照。样品用SDS、蛋白酶K、核酸酶处理后,用酚、苯酚/氯仿及氯仿/异戎醇抽提、处理纯化。取得的叶绿体DNA(ctDNA)用紫外分光光度计检测浓度与纯度,1%琼脂糖电泳得到清晰、整齐的条带。 The green leaves of vegetables through tissue homogenate, differential centrifugation separation, density gradient centrifugation purify sample of chloroplast. Examine the pure and shape of these chloroplast by microscope. The round and ellipse chloroplast was observed, then handle the chloroplast with SDS. DNaseI and proteinase K, after that extract the ctDNA with phenol/chloroform and chloroform/isoamyl alcohol. Examine this extractive of ctDNA with the 1% agarose gel electrophoresis and the quantity of with ultrauilet spectrophotometer,and the zone of the ctDAN is regular and clear.
