摘要
目的观察2型糖尿病(T2DM)大鼠逼尿肌细胞线粒体超微结构的改变。方法建立T2DM大鼠动物模型,利用透射电镜观察其逼尿肌细胞内线粒体结构并应用BI2000序列图像分析软件检测线粒体密度。结果对照组的线粒体相对光密度为2.417±0.376,相对灰度为0.572±0.083;T2DM组逼尿肌细胞内线粒体相对光密度为1.549±0.871,相对灰度为0.836±0.279,线粒体基质密度较正常明显下降。结论糖尿病大鼠逼尿肌细胞内线粒体超微结构破坏,基质密度下降,提示逼尿肌细胞能量代谢障碍是糖尿病膀胱收缩力降低的原因。
Objective To observe the ultrastructural changes of detrusor in experimental T2DM rats. Methods 14 Wistar rats were divided into two groups. The T2DM group rats underwent intraperitoneal inject with 30 mg/kg streptozotocin (STZ) 3 times, once a day. Then these rats were fed with the diets enriched with sucrose (20%, w/w), lard (18%, w/w) and yolk powder (3%, w/w) 2 weeks after they had been injected. Another group served as controls. The ultrastructures of mitochondrion in detrusor cells of each rats were examined after 12 weeks. Results The relative density of mitochondrion was 0.836±0.279 in T2DM group while 0.572±0.083 in the controls(P〈0.05). Conclusion Detrusor contraction of the bladder is impaired in the early stage of diabetes, the mitochondrion destroyed, and the detrusor metabolism dysfunction should been one of the causes.
