摘要 本实验采用SephadexG25及ACA凝胶柱层析技术,对神经肽注射液中多肽成份进行过柱分离。将过柱后收集的不同洗脱峰的溶液通过紫外扫描,茚三酮显色,微量双缩脲试验及薄层层析等技术进行鉴定。此外还用SDS—PAGE检测该注射液纯度及分子量。结果表明神经肽注射液含有一种主要多肽成份及大量小分子肽类,其主要多肽成份的分子量为14,000D。 The peptide components in neuropeptide injectio were separated with Sephadex G 25 and ACA gel filtration chromatography in this experiment. Then the liquid of different parts that were collected from gel column were identificated by UV scanning, ninhydrin, micro-- biuret reactions and thin--layer chromatography. Meanwhile, the purity and the molecular weight of the neuropeptide were determind by SDS -- Polyacrylamide gel electrophoresis, (SDS -- PAGE).The results showed that there were one main peptide component and some smaller molecular weight peptides in this injectic. The molecular weight of the main peptide component was about 14 , 400D.
