雌激素受体α和β在间充质干细胞体外成骨、成脂肪分化中的表达变化 被引量:6

摘要 目的:研究间充质干细胞(M SC)体外成骨、成脂肪分化与雌激素受体α和β表达变化的关系及其意义。方法:体外分离培养人M SC,流式细胞术鉴定其表型,加入成骨和成脂肪诱导剂,使用RT-PCR法检测骨桥蛋白(O steopon tin)和脂蛋白脂肪酶(lipoprote in lipase,LpL)的表达,半定量RT-PCR检测雌激素受体α(ERα)和β(ERβ)在成骨和成脂肪分化中的表达变化。结果:①体外分离培养的人M SC具有多向分化潜能,能向成骨细胞和脂肪细胞分化,分别表达O steopon tin和LpL。②M SC构成性表达雌激素受体α和β,雌激素受体α表达占优势。③在成骨分化中,雌激素受体α和β表达无明显改变,而在成脂肪分化中,雌激素受体α表达减低,β表达增高。结论:M SC成骨和成脂肪分化与雌激素受体α和β表达变化有关。调节雌激素受体α和β的表达,或使用一些雌激素受体激动剂、拮抗剂,将有可能抑制骨髓脂肪细胞的形成,促使M SC向成骨细胞分化,为骨质疏松及其它成骨障碍性疾病的治疗提供一个新的思路。
