摘要
本试验研究了用根癌土壤杆菌转化马铃薯的各种条件,在采用品种东农303的材料和A208、A281两种野生型根癌土壤杆菌的研究中筛选出了各个优化转化条件,建立了优化遗传转化系统.其结果为①外植体:块茎;②根癌土壤杆菌浓度:5×108CFU/ml;③pH值(共培养):5.8;④共培养中加入乙酰丁香酮浓度:40μm;⑤初始共培养时间:2天等的效果最佳.
Various conditions for potato (Solanum tubrosum L.) transformation with Agrobacterium tumefaciens A208 and A281 and cv. Dong Nong 303 were studied to construct optimized transformation system. The results were as follovos: ① explant, tuber; ② density of Agrobacterium tumefaciens, 5×108 CFU / ml; ③ pH value (co-cultivation), 5.8; ④ the concentration of added acctosyringonc, 40 μM; ⑤ co-cultivation time, 2 days.
