摘要
以马铃薯(Solanumtuberosum L.)"Wen-40"和"紫花白"2个品种的茎段和叶片为外植体,通过离体培养分化成苗以及对卡那霉素(Km)和头孢霉素(Cef)的敏感性研究,建立了农杆茵介导的稳定、高效的基因转化受体系统。结果表明:"Wen-40"的茎段适合转基因高效再生体系的建立,而25 mg.L-1的Km适合转基因植株的抗性筛选,低于200 mg.L-1的Cef浓度对马铃薯的诱导分化影响较小。
Stem segments and blades were used as explants of tissue culture of Solanum tuberosum L.varieties"Wen-40"and"Zihuabai",plantlet regeneration and effects of antibiotics,including kanamycin(Km),and cefotaxime(Cef),on the induction of callus were studied.The results indicated that stem segments was more suitable for establishment of regeneration system than blades.The optimized concentration of kanamycin for selecting the stem segments was 25 mg·L^-1 and cefotaxime was less than 200 mg·L^-1.
