摘要
用免疫组织化学方法,观察和计数了16周SD大鼠和衰老SD大鼠(24个月)脑血管周巨噬细胞。冷冻切片厚度为16μm。结果显示,特异性巨噬细胞单抗(ED2)染色仅限于血管周巨噬细胞。OX-42阳性的小胶质细胞与ED2阳性的细胞不同,FVIII-RA染色证实,血管内皮细胞也为ED2染色阴性。高倍显微镜下计数(每平方毫米脑组织内巨噬细胞个数)表明,衰老的SD大鼠脑血管周巨噬细胞(7.5±0.8)明显高于16周SD大鼠(2.4±0.6,P<0.01)。结果提示,衰老与脑血管周巨噬细胞的增加有关。
For study of the pathology of aging and cerebrovascular diseases,the perivascular macrophages in the brains of 16-week-old SD rats and 2-year-old SD rats were investigated and counted by using the method of immunohistochemistry. The cryostat sections were cut into the thick of 16μm,The results showed that the ED2-specific staining was restricted in a perivascular location, and no ED2 staining was found in the resident microglia or in the endothelial cells,which were identified by staining with the antibodies FVIII-RA and OX-42.The number of perivascular cells per spuare milimeter of highpower field was significantly greater in 2-year-old rats(7.5±0. 8)than in 16-week-old rats(2. 4±0.6,P<0. 01 ).The results suggest that the advanced age is associated with increased accumulation of macrophages.
