摘要
目的分析转化生长因子(TGF)-β1在人肝细胞性肝癌(HCC)及非癌组织中的表达,并探讨其与肿瘤发生、发展及预后的关系。方法采用免疫组织化学方法,分别检测30例HCC癌灶组及其自身对照的非癌组TGF-β1的表达,并以生物素标记的HBV-DNA探针检测肝癌组织中HBV-DNA,并分析TGF-β1表达与HBV复制及其临床病理学特征的关系。结果无论是原发性肝癌还是转移性肝癌,TGF-β1均呈较高表达,且组间未见显著差异;TGF-β1在肝癌癌灶及非癌组表达存在显著差别,其阳性率分别为83.3%和43.3%,肝癌组明显高于非癌组(P<0.01)。HCC的癌灶组中TGF-β1阳性表达与肿瘤分化程度显著相关(P<0.05);而与肿瘤直径、数目无关(P>0.05);HBV-DNA阳性肝癌组织中TGF-β1表达显著高于HBVDNA阴性组。结论TGF-β1在肝细胞性肝癌中过度表达,且与HCC的分化程度有关,可作为肝癌早期诊断及预后判断的标志。
Ojective To investigate the expression of transforming growth factor (TGF)-β1 in different parts of hepatocellular carcinoma (HCC) and explore the relationship between its expression and formation, development, and prognosis of HCC. Methods Using the immunohistochemistry, TGF-β1 expression was detected in 30 HCCs and their non-cancerous tissues. Liver HBV-DNA was detected by in situ molecular hybridization technique. The relationship was investigated between TGF-β1 expression and HBV replication or their clinical pathological characteristics. Results The stronger expression of TGF-β1 was found in whether primary or secondary liver cancerous tissues. The positive rate of TGF-β1 expression was 83.3% in HCC tissue, and 43.3 % in non-cancerous liver tissues, respectively. A significant difference was presented between them ( P 〈0.01). The positive expression of TGF-β1 in HCC was correlated to tumor differentiation, but neither to tumor size nor numbers ( P 〉 0.05). The level of TGF-β1 expression in HBV-DNA-positive HCC group was significantly higher than that in HBV-DNA-negative group. Conclusion TGF-β1 is overexpressed in HCC and associated with degree of differentiation. The abnormality of TGF-β1 could be used to be a marker for early diagnosis and prognosis of HCC.
