摘要
目的研究角膜移植排斥反应与Fas系统诱导的细胞凋亡之间的关系。方法将Wistar大鼠随机分为4组(A正常对照组;B自体移植组;C异种移植组;D异种移植CsA治疗组)。以豚鼠角膜为供体建立Wistar大鼠异种正位穿透性角膜移植动物模型。术后观察角膜植片的平均存活时间和排斥反应指数(RI);免疫组织化学方法检测植片组织Fas及其配体的表达;TUNEL法染色评定细胞凋亡指数(AI)。结果B组植片平均存活时间>30d,C组平均为(9·125±2·357)d,与B组相比存活时间明显缩短(P<0·01);D组植片平均存活时间(13·778±2·539)d,较C组显著延长(P<0·01)。术后3d,各组角膜植片RI、Fas、FasL及AI均开始升高,B组约持续7d左右即下降至对照组水平,而C、D2组则继续升高,约于术后14d达高峰,持续1周左右才开始逐渐下降。在相应时间点,D组各指标的变化水平均较C组低,差异有显著性意义(P<0·05或P<0·01)。结论角膜移植术后植片组织存在Fas及其配体分子的异常表达,由此引起的异常细胞凋亡可能是移植排斥反应发生的重要机制之一。
Objective To reseach the relationship between rejection of heteropenetrating keratoplasty and apoptosis induced bv Fas system. Methods Wistar rats were randomly assigned to four groups(A: control group; B: autokeratoplasty group: C: heterokeratoplasty group: D: CsA-treated heterokeratoplasty group) .Corneal grafts were transplanted orthotopieally from guinea pigs to Wistar rats. In grafts of postoperation, mean survival time and rejection index (RI) were observed. Expressions of Fas and Fas ligand(FasL) were detected by immunohistochemieal staining.Apoptosis indexes(AI) were determined by TUNEL.Results The mean survival time of group B was more than 30 days, of group C was (9. 125 ± 2. 357) days which was dramatieally shortened compared with group B(P 〈 0.01 ). The survival time of Group D was (13. 778 ± 2.539) days, which was longer than that of group C.After operation, RI,Fas, FasLand apoptosis index(AI) in four group began to increase from the 3rd day. In group B, these indexes increased only lasting seven days and then decreased to the level of group A. While to group C and D, they continune to increase and came to their picks at about 14th days respectively. These high levels lasted about one week and then decreased gradually. Each index of group C and group D was statistically higher than that of group B and group C' s was also higher than group D's(P〈0.05 or P〈0.01 ) .Conclusion After heterokeratoplasty, there are abnormal expressions of Fas and Fas ligand in grafts, which is probably one of important mechanisms of graft rejection.
